Screening of Salt Tolerant Potato Genotypes using Salt Stress and Molecular Markers

Aims: To screen potato genotypes for salt tolerance using in vitro stress and moleulcar markers.
 Experimental Design: The experiment was arranged Completely Randomized Design (CRD) with three replications.
 Place of the Study: conducted at Molecular Biology Tissue Culture Laboratories, Institute Biotechnology Genetic Engineering, Bangabandhu Sheikh Mujibur Rahman Agricultural University, Gazipur, Bangladesh.
 Methodology: In screening five (CIP 112, CIP 117, 120, 127, 128) done an agar medium tissue culture technique different concentrations viz. 0.0, 10, 20, 40, 60, 80, 100, 120 mM NaCl. All were further analysed through SSR markers primers. DNA bands visualized on gel electrophoresis scored polymorphic loci, gene diversity, genetic distance. NTSYSpc program used constructing unrooted neighbor-joining tree scatter diagram.
 Results: Potato 112 117 emerged as most tolerant highest plant height, shoot dry weight, root length, weight 127 128 poorly to stress. sensitive genotype produced minimum NaCl concentrations. Results indicated that significant differences found among cultivars. banding pattern microsatellite confirmed a distinct polymorphism tolerant, moderately lines. clustering suggests variations occur due genotypic variation possibly not epigenetic adaptation under conditions.
 Conclusion: pototo can be developing genotypes.
